Clozaril Information

The active ingredient in Clozaril is Clozapine. Other inactive ingredients may include:

• Lactose monohydrate: Approximately 100 mg – a filler and stabilizer.
• Microcrystalline cellulose: Approximately 50 mg – helps maintain tablet integrity.
• Magnesium stearate: Approximately 10 mg – a lubricant used in tablet manufacturing.
• Silicon dioxide: Approximately 5 mg – an anti-caking agent.

Common side effects include:

• Drowsiness or sedation
• Weight gain
• Constipation
• Drooling
• Dizziness or lightheadedness

Serious side effects that require immediate medical attention:

• Agranulocytosis (a severe reduction in white blood cells, which can lead to infection)
• Severe drowsiness or confusion
• Seizures
• High fever, muscle rigidity (Neuroleptic Malignant Syndrome)
• Signs of heart problems (irregular heartbeat, chest pain)

Clozaril is usually taken once or twice daily, with or without food. It is important to take the medication consistently at the same time each day to help maintain an even level of the drug in the bloodstream.

Important considerations:

• Regular blood tests are required to monitor white blood cell count, as Clozaril can cause a dangerous decrease in white blood cells (agranulocytosis).
• Do not stop taking Clozaril abruptly without consulting your doctor.
• Limit alcohol consumption, as it can increase drowsiness.

Clozaril can interact with other medications, so it’s important to inform your healthcare provider about any other drugs you are taking.

Common interactions:

• Other CNS depressants (e.g., alcohol, benzodiazepines): May increase sedation and respiratory depression.
• Anticholinergic drugs: May increase the risk of dry mouth, constipation, and blurred vision.
• Medications that affect the heart: May increase the risk of heart arrhythmias.
• Antidepressants or mood stabilizers: May alter the effectiveness of Clozaril or increase side effects.

Clozaril may take several weeks to show significant effects. While some improvements in symptoms may be seen within a few days to a week, full therapeutic effects may take up to 6 weeks or longer.

It is important to continue taking the medication even if you feel better, as sudden discontinuation can worsen symptoms.

If you experience severe side effects such as a high fever, muscle rigidity, confusion, or seizures, seek emergency medical attention immediately.

For signs of agranulocytosis (sore throat, fever, unusual weakness), contact your doctor as blood tests will be needed to assess your white blood cell count.
